Activity 11.2 Let us explore

Aim: To study the process of budding in yeast under a compound microscope.

Materials Required: Test tubes, conical flask, cotton plug, yeast granules, 10 g sugar, 100 mL water, slide, coverslip, dropper and microscope.

Procedure:

  1. Take 20 mL of sugar solution (1 g in 10 mL) in a test tube and add a pinch of yeast granules to it.
  2. Shake well and then put a cotton plug on the mouth of the test tube and keep it undisturbed in a warm place.
  3. After 1 or 2 hours, take out a small drop of yeast culture from the test tube with the help of a dropper on a slide and cover it with a coverslip.
  4. Observe the slide under a microscope at different magnifications.

Observation:

  1. At first a large number of small cells are seen, as you can see in figure (a).
  2. When seen under high power microscope, transparent and oval cells of yeast with buds are observed, as you can see in figure (b).

Conclusion: This shows that yeast multiplies by budding. The observations help us to understand that yeast multiplies by cell division (mitosis), producing genetically identical offspring rapidly under favourable conditions.
